Shinpei Tanaka: „Fluorescence correlation spectroscopy and Brownian dynamics simulation on confined protein molecules in lipid cubic phases”
University of Hiroshima, Japan

Protein solutions confined in lipid cubic phases are known to exhibit an enhanced crystallization behavior. To study this phenomenon, especially in the aspect of dynamics, we used the fluorescence correlation spectroscopy (FCS) to measure the diffusion coefficient of protein molecules confined in water channels of the cubic phases. Brownian dynamics simulations were also carried out to interpret the reduction of the diffusion coefficient obtained experimentally. In this talk, an explanation using a free-energy barrier for the confined diffusion and the importance of the hydrodynamic interaction will be presented.
